Detection of modified tyrosines as an inflammation marker in a photo-aged skin model

Reactive nitrogen species, produced during the process of inflammation induced by various factors including UV radiation, modify amino acids in crucial proteins. To investigate the influence of the modified tyrosine on UV-exposed skin, we detected the nitrotyrosine or halogenated tyrosine and dityrosine in photo-aged model mice. In this study, we used a low dose of UV irradiation to which we are exposed in daily life. Our results suggest that UV exposure in daily life may induce the production of modified tyrosines and skin aging.

Astaxanthin improves muscle lipid metabolism in exercise via inhibitory effect of oxidative CPT I modification

The present study investigated the effect of astaxanthin on muscle lipid metabolism in exercise. ICR mice (8 weeks old) were divided into four different groups: sedentary, sedentary treated with astaxanthin, running exercise, and exercise treated with astaxanthin. Our results suggested that astaxanthin promoted lipid metabolism rather than glucose utilization during exercise via CPT I activation, which led to improvement of endurance and efficient reduction of adipose tissue with training.

「光老化皮膚におけるチロシン修飾物の検出」 日本フリーラジカル学会/日本過酸化脂質フリーラジカル学会合同開催 2007年6月
紫外線は皮膚の肥厚や深いシワの形成をもたらすと考えられているがその直接的な関与についての報告は少ない。タンパク質を修飾する活性窒素種に着目し、その反応産物であるチロシン修飾物の検出を行ったところ、UV照射によりひきおこされた炎症の過程で修飾チロシンが生成された。今回のUV照射線量は日常レベルであることから、我々の結果は日常的な紫外線暴露が修飾チロシンの生成を誘導する可能性を示すものである。

「イソチオシアネート類とリジン残基の反応について」日本農芸化学会 2008年3月
ワサビの辛み成分であるアリルイソチオシアナートとリジン残基の反応について中性条件下で調べたところ、蛋白質SH基とも反応するが、リジン残基にも付加修飾することが化学的及び免疫化学的な手法により明らかとなった。

ω3系脂肪酸の酸化に伴う脳内蛋白質の修飾について調べたところ、タウ蛋白質がプロパノイル化されていることを免疫沈降法などにより明らかにした。

Inhibitory effect of phenolic antioxidants on eosinophil peroxidase

In this study, two methods for evaluation of inhibitory effects of eosinophil peroxidase (EPO) were established. Phenolic antioxidants strongly inhibited the formation of modified Tyr and the inhibitory effects increased with increasing phenolic OH moieties. Tocopherol, butylated hydroxytoluene (BHT), and cinnamic acid could not inhibit. Reduced and oxidized glutathione can inhibit the bromination but not dimerization of tyrosine, suggesting glutathione inhibit HOBr-derived modification but not HOBr generation itself from EPO.

Modification of lysine by isothiocyanates

The chemical reaction of ITC with bovine serum albumin (BSA) has been investigated. The amounts of lysine in acid hydrolysates of BSA were specifically reduced by ITC treatment. Next, Bz-Gly-Lys (Bz-GK) was reacted with ITC in buffers. New products from the reaction mixtures were detected and then identified as N-thiocarbamoyl-adducts by 1H-NMR and mass spectrometry. The adduct formation between Bz-Gly-Lys and grated Sawa-Wasabi was examined using LC/MS/MS. Both Allyl-ITC and 6-Methlsulfinylhexyl-adducts were detected in the mixture and the formation was increased with increasing amounts of Wasabi.

Preparation and characterization of a monoclonal antibody specific to allyl isothiocyanate-lysine adducts in proteins

When bovine serum albumin (BSA) was incubated with allyl isothiocyanate (AITC), considerable loss of Lys residues was observed. The antibody to AITC-protein was prepared and characterized. Using the antibody, we detected AITC-Lys from cultured cell by immunocytochemical staining. The formation of AITC-Lys in vivo following intake of Wasabi AITC is to be assessed.

To evaluate DHA or EPA-derived protein modification in vivo, we prepared and characterized a novel monoclonal antibody against propanoyl-lysine, PRL. The strong positive staining of atherosclerotic lesions from hypercholesterolemic rabbits was observed. In addition, specimens of old rat brain were more strongly stained by the antibody than these of young brain, suggesting enhancement of oxidative stress and/or failure of clearances of oxidatively modified biomolecules during brain aging.

Covalent modification of protein thiols by serotonin oxidation products formed by the catalytic action of myeloperoxidase

We examined the modification of thiols by serotonin in the presence of myeloperoxidase (MPO) and xanthine oxidase. In the presence of N-acetylcysteine (NAC), serotonin-dimer and three major NAC adducts were formed. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as a model thiol protein. Preliminary liquid chromatography mass spectrometry-based proteomic analysis indicated that one of adducts formed when TD reacted with the protein involved the peptide containing Cys-244. We propose that during inflammation MPO will oxidize serotonin resulting in covalent modification of thiol residues on proteins.

The data shown that iNOS mRNA expression and NO-modified protein in H.pylori-associated atrophic gastric mucosa were increased. These modified proteins might be involved in gastric carcinogenesis.

Novel biomarker of oxidative stress: oxidized omega-3 fatty acid-lysine adducts

In this study, we have reported that propanoyl-lysine (propionyl-lysine, PRL), as an oxidized omega-3 fatty acid-derived lysine adduct, was quantified from human urine by LC/MS/MS with isotopic dilution method. The amounts of urinary PRL and hexanoyl-lysine (HEL) in diabetic urines were higher than those of control human urines. Using the specific anti-PRL antibody, the presence in atherosclerotic plaque of cholesterol-fed rabbit has been confirmed. These lipid-lysine amide-type adducts family may be useful biomarkers for oxidative stress in vivo.
